All reagents used in a traditional sandwich ELISA are added in solution to a plate followed by a wash step and detection with the TMB colorimetric substrate.
SMAD1 is a member of a family of transcription factors involved in mediating signaling by growth and differentiation factors such as transforming growth factor beta (TGF-b) and bone morphogenic protein (BMP). SMAD1, along with SMADs 2, 3, 5, and 9, belongs to a class of SMAD proteins that is activated via phosphorylation by specific receptors in response to ligand binding. SMADs 1, 5, and 8 are activated by members of the BMP family. The canonical BMP signaling pathway involves SMAD phosphorylation at the SxS motif by activated BMP receptor kinase/ALK type I receptors. Phosphorylated SMAD1 associates with SMAD4, which is important for its function in regulating transcription. SMADs are involved in a range of biological activities including cell growth, differentiation, apoptosis, morphogenesis, development, and immune responses.
