Description: This FlowCytomix Simplex Kit is designed for the measurement of RANTES in an immunoassay analyzed on a flow cytometer. Together with the FlowCytomix Rat Basic Kit (cat. BMS8441FF), this kit can be used to detect RANTES alone or can be multiplexed with other Simplex Kits to measure a variety of analytes.
In this kit, the antibodies are mouse-specific and the standards are recombinant mouse proteins. The antibodies are cross-reactive with rat cytokines.
This kit contains bead population B4.
RANTES (Regulated upon Activation, Normal T cell Expressed and presumably Secreted), also known as CCL5, is a member of the CC chemokine familiy of inflammatory and immunoregulatory cytokines.
Similar to other CC chemokines RANTES is a potent chemoattractant for a number of different cell types including monocytes, unstimulated CD4+/CD45RO+ memory T-cells, stimulated CD4+ and CD8+ Tcells with na‹ve and memory phenotypes, NK cells, eosinophils, basophils, dendritic cells and mast cells. The activity of RANTES however is not restricted merely to chemotaxis, moreover it is a powerful activator for T-cells, monocytes, neutrophils and NK cells, a feature potentially relevant in a range of inflammatory disorders.
Several CC chemokine receptors, including CCR-1, CCR-3, CCR-4 and CCR-5, have been shown to bind RANTES and subsequently to transduce a signal by increasing the intracellular calcium ion level.
RANTES, amongst other chemokines, has been shown to play an important role in artherosclerosis, acute and chronic allergic inflammation, HIV suppression, food allergy and macrophage accumulation in the uterus during pregnancy. RANTES deficient mice showed impaired T-cell function.
