Description: Mouse Stem Cell Factor (SCF) is a hematopoietic growth factor and ligand for the KIT tyrosine kinase receptor, which exerts its activity in the early stages of hematopoiesis. Mouse SCF stimulates the proliferation of myeloid, erythroid, and lymphoid progenitors in bone marrow cultures.
Applications Reported: Recombinant mouse SCF is biologically active.
Applications Tested: The ED50 of this protein, as measured by MC/9 cell proliferation assay, is less than or equal to 65 ng/mL. This corresponds to a specific activity of greater than or equal to 1.5 x 10e4 Units/mg.
Source: E. coli expressed amino acids Lys 26-Ala 189 of mature mouse SCF accession # M59915.
Bioactivity: The ED50 of this protein, as measured by MC/9 cell proliferation assay, is less than or equal to 65 ng/mL. This corresponds to a specific activity of greater than or equal to 1.5 x 10e4 Units/mg.
Endotoxin: Less than 0.01 ng/ug cytokine as determined by the LAL assay. Purity: Greater than or equal to 95% as determined by SDS-PAGE.
Molecular Weight: The protein is not methionylated at the N-terminal and has a predicted molecular mass of 18,298. The DTT reduced protein migrates as an 18 kDa polypeptide on SDS-PAGE. The non-reduced protein migrates as a 16 kDa polypeptide.
Storage and handling: For best recovery, quick-spin vial prior to opening. Use in a sterile environment.
Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.
Stem cell factor (SCF) is the ligand of the c-Kit oncogene and is expressed by various structural and inflammatory cells in the airways. Binding of SCF by the c-Kit receptor leads to homodimerization of the receptor and the activation of signalling pathways such as PI-3, PLC-gamma, Jak/STAT, and MAP kinase pathways. SCF expression leads to the induction of mast cell survival and the expression and release of histamine, pro-inflammatory cytokines and chemokines. The inhibition of the SCF/c-Kit pathway leads to a decrease in histamine levels, mast cell and eosinophil infiltration, IL-4 production and airway hyperresponsiveness, suggesting this pathway may be a useful therapeutic target in inflammatory diseases such as asthma. At least two isoforms of SCF are known to exist.
