Description: Mouse CCL5, or RANTES (regulated on activation normal T cell expressed and secreted), is an 8 kDa monomeric protein that belongs to the CC- family of chemokines. The chemokines in this family share a common g-protein coupled receptor, CCR5, which they bind to through a conserved core region. In humans, CCR5 is also known to be the co-receptor through which the HIV virus enters the cell. This effect is blocked by the binding of RANTES to CCR5, making these proteins potential targets for HIV therapy.
RANTES plays an important role in the immune response by initiating the recruitment of leukocytes to the site of inflammation. It is a chemoattractant for T cells, monocytes, and dendritic cells, and may increase the cytotoxicity and promote proliferation of NK cells. It is reported to be produced by fibroblasts, platelets, and T cells. RANTES is also involved in the progression and metastasis of some forms of cancer, and constitutive expression has been demonstrated in some malignant tumors.
Human and mouse RANTES share 85% sequence homology and demonstrate cross-species activity.
Applications Reported: Recombinant mouse CCL5 (RANTES) is biologically active and can promote chemoattraction of mouse T cells in culture.
Applications Tested: This protein has been demonstrated to chemoattract murine T cells when used at 1.0-10.0 ng/mL.

Storage and handling: For best recovery, quick spin vial prior to opening. Use in sterile environment. Reconstitute in water to a concentration of 0.1-1.0 mg/mL. Do not vortex. Store lyophilized protein at less than or equal to -20°C. Reconstituted protein solution can be stored with carrier protein (e.g., 0.1% BSA) at less than or equal to -20°C.
RANTES is a chemoattractant for blood monocytes, memory T helper cells and eosinophils. It causes the release of histamine from basophils and activates eosinophils. It binds to CCR1, CCR3, CCR4 and CCR5 and is one of the major HIV suppressive factors produced by CD8+ T cells. Recombinant RANTES protein induces a dose dependent inhibition of different strains of HIV 1, HIV 2, and simian immunodeficiency virus (SIV). It is T cell and macrophage specific, induced by mitogens, and belongs to the intercrine beta (chemokine CC) family.
