The Human Tyrosine Kinase Receptor (Hu TIE-2) ELISA quantitates Hu TIE-2 in human serum, plasma, buffered solution, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu TIE-2.
Principle of the method
The Human TIE-2 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

The TEK receptor tyrosine kinase is expressed almost exclusively in endothelial cells. TEK signaling appears to be critical for endothelial cell-smooth muscle cell communication in venous morphogenesis, and defects in TEK are associated with inherited venous malformations. The ligand angiopoietin-1 binds to this receptor and mediates a signaling pathway that functions in embryonic vascular development. Immunoblotting showed that TIE2 expression was increased by thyroid-stimulating hormone and agents that increased intracellular cAMP. HSCs expressing the receptor tyrosine kinase TIE2 are quiescent and antiapoptotic and comprise a side population of HSCs that adhere to osteoblasts in the bone marrow niche.
