Description: The monoclonal antibody 1G5 recognizes human alpha catenin. Alpha catenin plays a role in cell-cell adhesion through its function in the binding and bundling of actin. The N-terminus of alpha catenin binds beta catenin and the C-terminus binds actin. Monomeric alpha catenin acts as a physical linker between the cadherin-beta catenin complex and the actin cytoskeleton and is stabilized by dimeric alpha catenin found in the cytoplasm. Alpha catenin also binds other actin-related proteins, including vinculin, alpha-actinin, ZO-1, afadin, formin-1, and Rho. Alpha catenin is absent in some tumor cell lines and reduced in some human carcinomas.
Applications Reported: This 1G5 antibody has been reported for use in flow cytometric analysis, immunoprecipitation, western blotting, immunohistochemical staining of formalin-fixed paraffin embedded tissue sections, and immunocytochemistry.
Applications Tested: This 1G5 antibody has been tested by immunohistochemistry of formalin-fixed paraffin embedded human tissue using high pH antigen retrieval or by immunocytochemistry of methanol- or formaldehyde-fixed and permeablized human cells and can be used at less than or equal to 5 µg/mL. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.
Alpha catenin is a member of the b-catenin family of cytosolic proteins. Alpha catenin interacts with N-cadherin and b-catenin on the cytosolic side of adherins junctions to mediate the interaction with the actin cytoskeleton. Alpha catenin is a homodimer in solution, but dissociates to form a 1:1 heterodimer with b-catenin in order to interact with cadherins. It also regulates morphological plasticity of synapses and cerebellar and hippocampal lamination during development. Alpha catenin also functions in the control of the startle modulation.
