Staining of normal human peripheral blood cells with APC anti-human CD19 (HIB19) (cat. 17-0199) and eFluor™ 605NC Mouse IgG1, Isotype Control (cat. 93-4714) (left) or eFluor^TM 605nm anti-human CD8 RPA-T8) (right). Cells in the lymphocyte gate were used for analysis.

Description

The RPA-T8 monoclonal antibody reacts with the human CD8a molecule, an approximately 32-34 kDa cell surface receptor expressed either as a heterodimer with the CD8 β chain (CD8 αβ) or as a homodimer (CD8 αα). A majority of thymocytes and a subpopulation of mature T cells and NK cells express CD8a. CD8 binds to MHC class I and through its association with protein tyrosine kinase p56lck plays a role in T-cell development and activation of mature T cells.

Applications Reported

For research use only, not for diagnostic or therapeutic use. This RPA-T8 antibody has been reported for use in flow cytometric analysis.

Applications Tested

This RPA-T8 antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells using eFluor^TM NC Flow Staining Buffer (Cat. 00-3222. This can be used at 5 μl per million cells in a 100 μl total staining volume.

The isotype control eFluor^TM 605^NC mouse IgG1 (cat. 93-4714) should be used at 5μL/test.

Laser/Filter Recommendation: When using eFluor^TM 605^NC, we recommend excitation with the 405nm violet laser with an appropriate filter set, such as the 595LP dichroic mirror with the 605/40 bandpass filter. An acceptable alternative is the 610/20 bandpass filter. Please contact eBioscience Technical Support for more information.

Buffer Recommendation: Comparison of eFluor^TM Nanocrystal conjugated antibody staining in different buffers has demonstrated that optimal performance is seen with the eFluor^TM NC Flow Cytometry Staining Buffer (cat. 00-3222). For a comparison of staining with different buffers, click here.

Whole Blood Recommendation: If staining whole blood samples, collection into heparin or sodium citrate-containing tubes is recommended. If EDTA is being used as an anti-coagulant, lysis of RBCs with eBioscience 1X RBC Lysis Buffer (cat. 00-4333) before staining is required.
