Mouse splenocytes were stimulated with ConA for 2 days, followed by IL-2 and IL-4 for 3 days, and restimulated with immobilized anti-CD3 and soluble anti-CD28 in the presence of Brefeldin A for 5 hours. The cells were fixed and permeabilized and subsequently stained with 0.125 ug Pacific Blue anti-mouse TNF-a (MP6-XT22) (right) or staining buffer (autofluoresence) (left).

Description


The MP6-XT22 antibody reacts with mouse tumor necrosis factor-alpha (TNF-α), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells and CD4+ T cells. TNF-α has cytolytic activity against a range of tumor cells and is important in immune regulation. TNF-α forms dimers and trimers and also exists as a 26 kDa membrane-bound form.


Applications Reported


For research use only, not for diagnostic or therapeutic use. This MP6-XT22 antibody has been reported for use in intracellular staining followed by flow cytometric analysis.

The Pacific Blue® emits at 456 nm and can be excited with the Violet laser. Please make sure that your instrument is capable of detecting this fluorochome.


Applications Tested


This MP6-XT22 antibody has been tested by intracellular staining and flow cytometric analysis of mouse splenocytes. This can be used at less than or equal to 0.25 μg per million cells in a 100 μl total staining volume. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
