Human PBMCs were primed with IFNγ for 2 hrs, then activated with LPS for 22 hrs in the presence of monensin. Cells were then intracellularly stained with staining buffer (autofluorescence) (left) or Pacific Blue® anti-human IL-12 p40/70 (C8.6) (right). Cells in the monocyte gate were used for analysis.

Description


The C8.6 antibody reacts with the p40 subunit of human IL-12 p70 and human IL-23. IL-12 is a 70 kDa heterodimeric cytokine composed of two covalently linked chains, p40 and p35, and secreted primarily by activated monocytes, macrophages, and dendritic cells. IL-23 is a biologically-related, heterodimeric cytokine composed of disulfide-linked p40 and p19 subunits.


Applications Reported


For research use only, not for diagnostic or therapeutic use. The C8.6 antibody has been reported for use in ELISA, neutralization, and intracellular staining for flow cytometric analysis.


Applications Tested


This C8.6 antibody has been pre-titrated and tested by intracellular staining and flow cytometric analysis. This can be used at 20 μl (0.125 μg) per 100 μl blood (or per 1 million cells in 100 μl total staining volume).
