Normal human peripheral blood mononuclear cells were restimulated with PMA and Ionomycin in the presence of monensin for 5 hours and then stained intracellularly with FITC anti-human CD4 (RPA-T4) (cat. 11-0049) and 0.03 μg of Alexa FluorŽ 647 Rat IgG2a Iso Cntrl (cat. 51-4321) (left) or 20µl of Alexa FluorŽ 647 anti-human GM-CSF (BVD2-21) (right). Total viable cells were used for analysis.

Description


The BVD2-21C11 monoclonal antibody reacts with human Granulocyte/Monocyte-Colony Stimulating Factor (GM-CSF). Human GM-CSF is a 127 amino acid, differentially glycosylated factor produced mainly by activated T cells and macrophages. Endothelial cells and fibroblasts can also produce GM-CSF after exposure to TNF-α, IL-1, IL-2 and IFN-γ. GM-CSF is found associated with the extracellular matrix and in membrane-bound formats as well. It stimulates proliferation, activation and differentiation of macrophages, granulocytes and their progenitors.


Applications Reported


For research use only, not for diagnostic or therapeutic use. This BVD2-21C11 antibody has been reported for use in intracellular staining followed by flow cytometric analysis.


Applications Tested


This BVD2-21C11 antibody has been pre-titrated and tested on peripheral blood mononuclear cells reactivated with PMA and Ionomycin in the presence of monensin. This can be used at 20 μl (0.03 μg) per million cells in a 100 μl total staining volume.
