BALB/c splenocytes were cultured for 3 days with plate-bound anti-mouse CD3e (clone 145-2C11, cat. 16-0031), soluble anti-mouse CD28 (clone 37.51, cat. 16-0281), recombinant mouse IL-2 (cat. 14-8021), and recombinant mouse IL-4 (cat. 14-8041). Cells were then harvested and restimulated with PMA/Ionomycin in the presence of monensin for 5 hours. Following restimulation, cells were harvested, surface stained with FITC anti-mouse CD4 (clone RM4-5, cat. 11-0042), fixed and permeabilized, then stained with 0.06 μg Alexa Fluor 647 eBio13A (left). The ligand blocking control demonstrates that pre-blocking the Alexa Fluor 647 conjugated eBio13A antibody with 1μg of recombinant mouse IL-13 (cat. 14-8131) prior to staining abrogates specific mouse IL-13 staining (right).

Description


The eBio13A antibody reacts with mouse IL-13. IL-13 is a cytokine produced mainly by Th2 cells, but also by antigen-primed CD8 T cells. IL-13 has a strong involvement in allergic inflammation and parasitic clearing and in cancer models has been shown to have either inhibitory or stimulatory activity depending on the tumor. In humans, IL-13 is found to play a role in isotype switching in B cells. IL-13 is implicating in down modulating macrophage activity, through the reduction of pro-inflammatory cytokines (IL-1, IL-6, IL-8, IL-10, IL-12)




Applications Reported


For research use only, not for diagnostic or therapeutic use. The eBio13A antibody has been reported useful for ELISA and ELISPOT capture, as well as intracellular staining for flow cytometric analysis.


Applications Tested


This eBio13A antibody has been tested by intracellular staining and flow cytometric analysis of mouse splenocytes. This can be used at less than or equal to 0.125 μg per million cells in a 100 μl total staining volume. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
