Description


The monoclonal antibody eBio4E5 reacts with Ly49D, an activating member of the Ly49 family of NK cell receptors. The receptors form homodimers which bind MHC Class I alloantigens. The activating Ly49 receptors (Ly49D and Ly49H) do not contain any immune receptor tyrosine-based inhibitory motif (ITAM) in their cytoplasmic domains unlike inhibitory Ly49 family members. Alternatively, Ly49 molecules have been shown to associate with a 12-kDa homodimeric protein, DAP12, that contains an immunoreceptor tyrosine-based activation motif critical for positive signaling by these receptors. Ly49D can mobilize intracellular 2+Ca and mediate reverse antibody-dependent cellular cytotoxicity in the presence of specific antibody. Expression of family members is independent therefore several Ly49 proteins can be expressed on the same cells. Expression of Ly49D is strain dependent: on subsets of natural killer (NK) cells in C57BL/6, CB6F-1 and SJL, while BALB/c, DBA/2, AKR, and CBA/J are negative. In 129/J mice, eBio4E5 also recognizes Ly49O and Ly49V and LY49R.

Applications Tested


This eBio4E5 (4E5) antibody has been tested by flow cytometric analysis of C57Bl/6 mouse splenocytes. This can be used at less than or equal to 0.5 μg per million cells in a 100 μl total staining volume. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
