Description
Mouse IFN-γ is a ~20 kDa factor produced by activated T, B and NK cells, and is an anti-viral and anti-parasitic cytokine. IFN-γ, in synergy with other cytokines such as TNF-α, inhibits proliferation of normal and transformed cells. Immunomodulatory effects of IFN-γ are exerted on a wide range of cell types expressing the high affinity receptors for IFN-γ. Glycosylation of IFN-γ does not affect its biological activity.
Applications Reported
For research use only, not for diagnostic or therapeutic use. Recombinant mouse IFN-γ is biologically active. The intended use of the Recombinant Mouse IFN-γ Single-Use ELISA RSG Standard is as a standard for mouse IFN-γ Ready-SET-Go! ELISA and mouse IFN-γ matched antibody pair ELISA. Typically the standard is run in a series of 7 doubling dilutions from the ‘top’ standard described below.
Instructions for use (XMG1.2/R4-6A2 assays):
For Ready-SET-Go! ELISA Set (TMB substrate). Dilution: Add 20 ul of standard solution to 10 ml of assay diluent to prepare the top standard solution (2000 pg/ml).
For matched antibody pair ELISA (ABTS or Super AquaBlue substrate). Dilution: Add 20 ul of standard solution to 10 ml of assay diluent to prepare the top standard solution (2000 pg/ml).
Instructions for use (AN-18/R4-6A2 assays):
For Ready-SET-Go! ELISA Set (TMB substrate). Dilution: Add 20 ul of standard solution to 10 ml of assay diluent to prepare 2000 pg/ml solution. Mix thoroughly. Further dilute by adding 50 ul of this solution to 0.95 ml of assay diluent to make the top standard solution (100 pg/ml).
For matched antibody pair ELISA (ABTS or Super AquaBlue substrate). Dilution: Add 20 ul of standard solution to 10 ml of assay diluent to prepare 2000 pg/ml solution. Mix thoroughly. Further dilute by adding 100 ul of this solution to 0.90 ml of assay diluent to make the top standard solution (200 pg/ml).
Note: These are single-use vials. Use one time and discard. Dilute as suggested above, not difficult-to-measure fractions of the suggested volumes (e.g., not 0.5 ul in 1.0 ml) or recovery may not be accurate. Use within 6 hours of dilution to top standard concentration.
Applications Tested
This cytokine standard has been calibrated, and subsequently tested by sandwich ELISA using the mouse IFN-γ ELISA pair (XMG1.2/R4-6A2 or AN-18/R4-6A2). These two different assays have different dynamic ranges.
