Description
The AIO.3 antibody reacts with mouse IL-15. This is a neutralizing antibody. Mouse IL-15 is a 14-15 kDa factor, previously known as IL-T. IL-15 is produced by a variety of cells and has a role in the T cell-mediated immune response and the central nervous system. IL-15 binds to the β and γ subunits of the IL-2 receptor; however, these two cytokines have distinct biological activities. IL-15 is a specific maturation factor for NK cells and mimics the BM microenvironment for NK cell development in vitro. It is reported that IL-15 delivers an anti-apoptotic and survival signal for NK, T, and B cells.

The Functional Grade Biotin anti-mouse IL-15 antibody (clone AIO.3) is available for beta test use as the in vivo capture antibody for the In Vivo Cytokine Capture Assay (IVCCA). eBioscience is making this reagent available in a collaborative effort to help speed development and optimization of this assay. A reagent credit is offered to collaborating laboratories which further validate the mouse IL-15 IVCCA using these reagents.
For 6 different cytokine assays previously described, the IVCCA has been found to facilitate measurement of cytokines in serum by increasing their in vivo half lives dramatically. This increases the sensitivity of measurement of in vivo cytokine production 30- to 1000- fold.

Applications Tested
The Functional Grade Biotin AIO.3 antibody has been tested by LAL assay to verify low endotoxin level, and it has been tested as the detection antibody in a sandwich ELISA for analysis of mouse IL-15 protein levels in combination with the purified anti-mouse IL-15 antibody for capture and recombinant mouse IL-15 (14-8151) as the standard.
A suitable range of concentrations of the FG Biotin AIO.3 antibody for ELISA detection is 1.0-4 µg/ml. A standard curve consisting of doubling dilutions of the recombinant standard over the range of 4000 pg/ml - 30.0 pg/ml should be included in each ELISA plate.
