Formulation: Sterile liquid; 1 mM citrate buffer, pH 3.5, with 0.1 % BSA as a stabilizer. 0.22 µm filtered.
Storage Conditions: For greatest stability, keep concentration of primary stock at or above 10 µg/ml. For long term storage, aliquot into polypropylene vials (volumes of 20 µl or greater) and store at or below -80°C. Avoid repeated freeze/thaw cycles.
Handling Conditions: For best recovery, always quick-spin vial prior to opening. For dilution of current stock, always include carrier protein (1% BSA or 10% FBS) in the buffered saline diluent.
Source: Human TGFb1 was expressed in insect cells. Mature TGFb1 (ala279 - ser390; accession #BC001180) was purified from the conditioned media.
Molecular Mass: Mature TGFb1 polypeptide has a predicted molecular mass of 12,794. On reducing SDS-PAGE the protein migrates as a 13 kDa polypeptide and on non-reducing SDS page the cystine-linked homodimer migrates as a 25 kDa protein.
Purity: Greater than 98%, as determined by SDS-PAGE
Endotoxin Level: Less than 0.01 ng/ug cytokine as determined by the LAL assay.
Bioactivity: Measured by inhibition of IL-4 induced mouse HT-2 cell proliferation. The ED50 is 0.3 ng/ml, corresponding to a specific activity of 3 x 106 Units/mg.

Description
Human TGF-β1 (Transforming Growth Factor-β1) is a member of a superfamily of homologous, disulfide-linked, homodimeric proteins that regulate the proliferation and differentiation of normal and transformed cells. Human TGF-β1 is a 25 kDa protein with each subunit containing 112 aa residues.
Applications Reported
For research use only, not for diagnostic or therapeutic use. Recombinant human TGF-β1 is biologically active and can inhibit IL-4 induced proliferation of mouse HT-2 cells. Recombinant human TGF-β1 is useful as an ELISA standard.
Applications Tested
This recombinant human TGF-β1 has been tested in bioassay for inhibition of IL-4-dependent proliferation of mouse HT-2 cells. The ED
50 measured in such assays is typically < 0.30 ng/ml, corresponding to a bioactivity of > 3 x 10
6 units/mg.
