Formulation: Sterile liquid; phosphate buffered saline, pH 7.2, 150 mM NaCl, with no carriers or preservatives. 0.22 µm filtered.
Storage Conditions: For greatest stability, keep concentration of primary stock at or above 10 µg/ml. For long term storage, aliquot into polypropylene vials (volumes of 20 µl or greater) and store at or below -80°C. Avoid repeated freeze/thaw cycles.
Handling Conditions: For best recovery, always quick-spin vial prior to opening. For dilution of current stock, always include carrier protein (1% BSA or 10% FBS) in the buffered saline diluent.
Source: E. coli expressed amino acids Gln 24-Gln 166 of mature human IFN-γ (accession # NM_000619).
Molecular Mass: The methionylated N-terminal protein migrates as a 16 kDa polypeptide on SDS-PAGE.
Purity: Greater than 98%, as determined by SDS-PAGE
Endotoxin Level: Less than 0.01 ng/ug cytokine as determined by the LAL assay.
Bioactivity: Measured by human A549/EMCV cell protection assay. The ED50 is 150 pg/ml, corresponding to a specific activity of 6.6 x 10E6 Units/mg.

Description


Human IFN-γ is an ~20 kDa factor produced by activated T, B and NK cells and is an anti-viral and anti-parasitic cytokine. IFN-γ in synergy with other cytokines, such as TNF-α, inhibits proliferation of normal and transformed cells. Immunomodulatory effects of IFN-γ are exerted on a wide range of cell types expressing the high affinity receptors for IFN-γ. Glycosylation of IFN-γ does not affect its biological activity.


Applications Reported


For research use only, not for diagnostic or therapeutic use. Recombinant human IFN-γ is biologically active. It has been demonstrated to protect A549 cell line against viral infection in culture. It is also useful as a cytokine standard in a human IFN-γ ELISA.


Applications Tested


This recombinant human IFN-γ has been tested by sandwich ELISA using the anti-human IFN-γ antibody pair MD-1/4S.B3 or the pair NIB42/4S.B3. A standard curve consisting of doubling dilutions of the recombinant standard over the range of 1000 pg/ml - 8 pg/ml should be included in each ELISA plate. It has also been tested in bioassays using mouse cell line A549. The ED50 measured in A549 protection is typically 300 pg/ml, corresponding to a specific activity of approximately 3.3x106 Units/mg.
