Product Information
Contents: Carrier-Free Recombinant Human IL-10 (Interleukin-10)
Catalog Number: 34-8109
Sizes: 100 ug, 500 ug
Formulation: Sterile liquid; phosphate buffered saline, pH 7.2, 150 mM NaCl, with no preservatives or carriers. 0.22 µm filtered.
Storage Conditions: For greatest stability, keep concentration of primary stock at or above 10 µg/ml. For long term storage, aliquot into polypropylene vials (volumes of 20 µl or greater) and store at or below -80°C. Avoid repeated freeze/thaw cycles.
Handling Conditions: For best recovery, always quick-spin vial prior to opening. For dilution of current stock, always include carrier protein (1% BSA or 10% FBS) in the buffered saline diluent.
Source: E. coli expressed amino acids Ser 19-Asn 178 of mature human IL-10 (accession # NM_000572).
Molecular Mass: The protein is not methionylated at the N-terminal and has a predicted molecular mass of 18,647. The DTT reduced protein migrates as an 18 kDa polypeptide on SDS-PAGE and the non-reduced protein migrates with slightly greater mobility.
Purity: Greater than 98%, as determined by SDS-PAGE
Endotoxin Level: Less than 0.01 ng/ug cytokine as determined by the LAL assay.
Bioactivity: Measured by mouse MC/9 cell proliferation assay, costimulated with IL-4. The ED50 is 1.0 ng/ml, corresponding to a specific activity of 1.0 x 10E6 Units/mg.

Description
Human IL-10 is a ~18 kDa factor also known as cytokine synthesis inhibitory factor (CSIF). Th2 T cells, B1 B cells and keratinocytes are the major cell subsets that produce IL-10. IL-10 inhibits synthesis of Th1 cytokines and proliferation of T cells and acts as a costimulatory signal for mast cells, developing thymocytes and Th2 response.
Applications Reported
For research use only, not for diagnostic or therapeutic use. Recombinant human IL-10 is biologically active and can promote proliferation of MC/9 cell line in culture. It is also used as standard in human IL-10 ELISA.
Applications Tested
This recombinant human IL-10 has been tested by sandwich ELISA using anti-human IL-10 antibody pair (JES3-9D7/JES3-12G8) and in bioassay using mouse cell line MC/9. The ED
50 measured in a MC/9 proliferation assay is typically 1.0 ng/ml, corresponding to a specific activity of approximately 1x10
6 Units/mg.
