Contents: Carrier-Free Recombinant Human IL-4 (Interleukin-4)
Catalog Number: 34-8049
Formulation: Sterile liquid; phosphate buffered saline, pH 7.2, 150 mM NaCl, with no carriers or preservatives. 0.22 µm filtered.
Storage Conditions: For greatest stability, keep concentration of primary stock at or above 10 µg/ml. For long term storage, aliquot into polypropylene vials (volumes of 20 µl or greater) and store at or below -80°C. Avoid repeated freeze/thaw cycles.
Handling Conditions: For best recovery, always quick-spin vial prior to opening. For dilution of current stock, always include carrier protein (1% BSA or 10% FBS) in the buffered saline diluent.
Source: E. coli expressed amino acids His 25-Ser 153 of mature human IL-4 (accession # AF 395008).
Molecular Mass: The protein is not methionylated at the N-terminal and has a predicted molecular mass of 14,963. The DTT reduced protein migrates as a 15 kDa polypeptide on SDS-PAGE and the non-reduced protein migrates with slightly greater mobility.
Purity: Greater than 98%, as determined by SDS-PAGE
Endotoxin Level: Less than 0.01 ng/ug cytokine as determined by the LAL assay.
Bioactivity: Measured by human TF-1 cell proliferation assay. The ED50 is 0.1 ng/ml, corresponding to a specific activity of 1.0 x 10E7 Units/mg.

Description
Human IL-4 is a ~15-19 kDa factor also known as BCDF, BCGF-1 and BSF-1. IL-4 is secreted by activated Th2 and natural killer T cells and to a lesser extent by Th1 and mast cells. IL-4 is species-specific and promotes proliferation and differentiation of B cells. IL-4 in conjunction with other signals such as CD40 induces Ig isotype switching of B cells and upregulation of surface antigens including MHC class II and CD23.
Applications Reported
For research use only, not for diagnostic or therapeutic use. Recombinant human IL-4 is biologically active and can promote proliferation of B lymphocytes in culture. It is also used as standard cytokine in human IL-4 ELISA.
Applications Tested
This recombinant human IL-4 has been tested by sandwich ELISA using anti-human IL-4 antibody pair (8D4-8/MP4-25D2) and in bioassays using human cell line TF-1. The ED
50 measured in a TF-1 proliferation assay is typically 100 pg/ml, corresponding to a specific activity of approximately 1x10
7 Units/mg.
