Formulation: Sterile liquid; 5 mM citric acid, 5 mM NaHPO4, 0.4 M NaCl, pH 4.0, 1 mM DTT, with no carriers or preservatives. 0.22 uM filtered.
Storage Conditions: For greatest stability, keep concentration of primary stock at or above 10 µg/ml. For long term storage, aliquot into polypropylene vials (volumes of 20 µl or greater) and store at or below -80°C. Avoid repeated freeze/thaw cycles.
Handling Conditions: For best recovery, always quick-spin vial prior to opening. For dilution of current stock, always include carrier protein (1% BSA or 10% FBS) in the buffered saline diluent.
Source: E. coli expressed amino acids Ala 21-Gln 169 of mouse IL-2 (accession # MMIL04).
Molecular Mass: The protein is a mixture of N-terminally methionylated and non-methionylated protein. Th DTT-reduced and non-reduced protein migrates as 18 kDa polypeptides on SDS-PAGE.
Purity: Greater than 98%, as determined by SDS-PAGE
Endotoxin Level: Less than 0.01 ng/ug cytokine as determined by the LAL assay.
Bioactivity: Measured by mouse CTLL2 cell proliferation assay. The ED50 is 0.1 ng/ml, corresponding to a specific activity of 1.0 x 10E7 Units/mg.

Description


Mouse IL-2 (also known as TCGF) is an ~17 kDa factor produced mainly by activated CD4+ T cells. IL-2 induces cell cycle progression of resting cells in an antigen non-specific manner and allows clonal expansion of activated T cells. IL-2 also acts on activated B cells, monocytes, NK cells, LAK cells, and on oligodendroglial cells in vitro. In addition, IL-2 plays a role in hematopoiesis, tumor surveillance and anti-inflammatory reactions, and hence is a central regulator of the immune response. Non-glycosylated IL-2 is biologically active.


Applications Reported


For research use only, not for diagnostic or therapeutic use. Recombinant mouse IL-2 is biologically active and can promote proliferation of mouse T lymphocytes in culture. It is also used as a standard in mouse IL-2 ELISA.


Applications Tested


This reagent has been tested by sandwich ELISA using anti-mouse IL-2 antibody pair (JES6-1A12/JES6-5H4) and in bioassays using the mouse cytotoxic T cell line CTLL-2. The ED50 measured in a CTLL-2 proliferation assay is typically 100 pg/ml, corresponding to a specific activity of (1 x 107 U/mg).
