Product Information
Contents: Carrier-Free Recombinant Human IL-1b (Interleukin-1 beta)
Catalog Number: 34-8018
Sizes: 100 ug, 500 ug
Formulation: Sterile liquid; phosphate buffered saline, pH 7.2, 150 mM NaCl, with no carriers or preservatives. 0.22 µm filtered.
Storage Conditions: For greatest stability, keep concentration of primary stock at or above 10 µg/ml. For long term storage, aliquot into polypropylene vials (volumes of 20 µl or greater) and store at or below -80°C. Avoid repeated freeze/thaw cycles.
Handling Conditions: For best recovery, always quick-spin vial prior to opening. For dilution of current stock, always include carrier protein (1% BSA or 10% FBS) in the buffered saline diluent.
Source: E. coli expressed amino acids Ala 117-Ser 269 of human IL-1b (accession # NM_000576).
Molecular Mass: The protein is not methionylated at the N-terminal and has a predicted molecular mass of 17,377. The DTT reduced and non-reduced proteins migrate as 18 kDa polypeptides on SDS-PAGE.
Purity: Greater than 98%, as determined by SDS-PAGE
Endotoxin Level: Less than 0.01 ng/ug cytokine as determined by the LAL assay.
Bioactivity: Measured by mouse D10S cell proliferation assay. The ED50 is 0.1 ng/ml, corresponding to a specific activity of 1.0 x 10E7 U/mg.

Description
Human IL-1β, also called lymphocyte activating factor (LAF), endogenous pyrogen (EP), leucocyte endogenous mediator (LEM), mononuclear cell factor (MCF), is produced by a wide variety of cells, including macrophages, dendritic cells, T cells and B cells. IL-1β is mostly released and has only 20% amino acid homology with IL-1α. The immune regulatory role of IL-1β is exerted on a wide range of cells including lymphocytes, epithelial cells and fibroblasts.
In vivo, it induces hypotension, fever, and acute phase response.
Applications Reported
For research use only, not for diagnostic or therapeutic use. Recombinant human IL-1β is biologically active. The protein is reported for use in bioassay and as the standard for ELISA.
Applications Tested
This reagent has been tested by sandwich ELISA using the anti-human IL-1β ELISA pair (CRM56/CRM57), and in bioassays, with an observed ED
50 of 100 pg/ml, corresponding to a specific activity of approximately 1x10
7 Units/mg.
