Description


The RB6-8C5 monoclonal antibody reacts with mouse Ly-6G and Ly-6C, a 21-25 kDa protein also known as the myeloid differentiation antigen Gr-1. A GPI-linked protein, Gr-1 is expressed by the myeloid lineage in a developmentally regulated manner in the bone marrow. While monocytes only express Gr-1 transiently during their bone marrow development, the expression of Gr-1 on bone marrow granulocytes as well as on peripheral neutrophils is a good marker for these populations.


Applications Reported


For research use only, not for diagnostic or therapeutic use. This RB6-8C5 antibody has been reported for use in flow cytometric analysis.

The photo-emission properties and compensation required for Alexa FluorŽ 750 are similar to APC-Cy7; however Alexa FluorŽ 750 has several advantages when compared to APC-Cy7. The Alexa FluorŽ 750 has peak emission of 779 nm, and usually has a slightly higher mean fluorescent intensity when compared to the same clone conjugated with APC-Cy7, which has peak emission of 767 nm. Furthermore, Alexa FluorŽ 750 is considered to be a more photostable fluorochrome as compared with APC-Cy7. It is advised not to use fixatives with Alexa FluorŽ 750 as this may lead to degradation of the fluorochrome and loss of staining.


Applications Tested


This RB6-8C5 antibody has been tested by flow cytometric analysis of mouse bone marrow cells and splenocyte suspensions. This can be used at less than or equal to 0.5 μg per million cells in a 100 μl total staining volume. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
