Staining of normal human peripheral blood cells with APC anti-human CD8a (RPA-T8) (cat. 17-0088) (FL5-H) and staining buffer (autofluorescence) (left) or APC-Alexa Fluor® 750 anti-human CD4 (RPA-T4) (right). Cells in the lymphocyte gate were used for analysis.

Description

The RPA-T4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface receptor expressed by a majority of thymocytes, subpopulation of mature T cells (T-helper cells) and in low levels on monocytes. CD4 is a receptor for the human immunodeficiency virus (HIV). RPA-T4 blocks HIV binding and mixed lymphocyte reaction. The RPA-T4 antibody recognizes a different epitope than the OKT4 monoclonal antibody, and these antibodies do not cross-block binding to each other's respective epitopes.

Applications Reported

For research use only, not for diagnostic or therapeutic use. This RPA-T4 antibody has been reported for use in flow cytometric analysis.

The photo-emission properties and compensation required for Alexa Fluor® 750 are similar to APC-Cy7; however Alexa Fluor® 750 has several advantages when compared to APC-Cy7. The Alexa Fluor® 750 has peak emission of 779 nm, and usually has a slightly higher mean fluorescent intensity when compared to the same clone conjugated with APC-Cy7, which has peak emission of 767 nm. Furthermore, Alexa Fluor® 750 is considered to be a more photostable fluorochrome as compared with APC-Cy7. It is advised not to use fixatives with Alexa Fluor® 750 as this may lead to degradation of the fluorochrome and loss of staining.

Applications Tested

This RPA-T4 antibody has been pre-titrated and tested by flow cytometric analysis of human peripheral blood leukocytes. This can be used at 20 μl (0.5 μg) per 100 μl blood (or per 1 million cells in 100 μl total staining volume).
