Description
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. Along with the other CD3 subunits γ and δ, the ε chain is required for proper assembly, trafficking and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Crosslinking of TCR via immobilized UCHT1 initiates an intracellular biochemical pathway resulting in cellular activation and proliferation.
Applications Reported
For research use only, not for diagnostic or therapeutic use. This UCHT1 antibody has been reported for use in flow cytometric analysis.
The photo-emission properties and compensation required for Alexa FluorŽ 750 are similar to APC-Cy7; however Alexa FluorŽ 750 has several advantages when compared to APC-Cy7. The Alexa FluorŽ 750 has peak emission of 779 nm, and usually has a slightly higher mean fluorescent intensity when compared to the same clone conjugated with APC-Cy7, which has peak emission of 767 nm. Furthermore, Alexa FluorŽ 750 is considered to be a more photostable fluorochrome as compared with APC-Cy7. It is advised not to use fixatives with Alexa FluorŽ 750 as this may lead to degradation of the fluorochrome and loss of staining.
Applications Tested
This UCHT1 antibody has been pre-titrated and tested by flow cytometric analysis of human peripheral blood leukocytes. This can be used at 20 μl (0.5 μg) per 100 μl blood (or per 1 million cells in 100 μl total staining volume).
