Mouse splenocytes were stimulated with ConA for 2 days, followed by IL-2 and IL-4 for 3 days, and restimulated with immobilized anti-CD3 and soluble anti-CD28 in the presence of Brefeldin A for 5 hours. The cells were surface stained with FITC anti-mouse CD4 (GK1.5) and intracellularly stained with PE anti-mouse TNF-a (MP6-Xt22). (Right) The cells were surface stained with PE anti-mouse CD4 and intracellularly stained with APC anti-mouse TNF-a.

Description
The MP6-XT22 antibody reacts with mouse tumor necrosis factor-alpha (TNF-α), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells and CD4
+ T cells. TNF-α has cytolytic activity against a range of tumor cells and is important in immune regulation. TNF-α forms dimers and trimers and also exists as a 26 kDa membrane-bound form.
Applications Reported
For research use only, not for diagnostic or therapeutic use. The MP6-XT22 antibody has been reported for use in intracellular flow cytometric analysis.
This tandem dye is sensitive to photo-induced oxidation. Protect this vial
from light during storage and all handling steps. When staining cells with
PE-Cy7-conjugated antibodies, always protect samples from light.
This product is guaranteed for 6 months upon receipt when stored properly.
Applications Tested
The MP6-XT22 antibody is offered in 2 formats:
- μg size: has been tested by intracellular flow cytometric analysis. This can be used at less than or equal to 0.5 μg per million cells in a 100 μl total staining volume. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
- test size: has been pre-titrated and tested by intracellular flow cytometric analysis. This can be used at 20 μl per million cells in a 100 μl total staining volume. The pre-titrated test size contains 0.2% BSA.
