Human peripheral blood mononuclear cells were stimulated with PMA and ionomycin in the presence of Brefeldin A for 5 hours. Subsequently, the cells were surface stained with FITC anti-human CD4 (OKT4) and intracellularly stained with PE-Cy7 mouse IgG1 isotype control (left) or PE-Cy7 anti-human IL-4 (8D4-8) (right).

Description
The 8D4-8 antibody reacts with human interleukin-4 (IL-4), a 15-19 kDa cytokine secreted by Th2 cells.
Applications Reported
For research use only, not for diagnostic or therapeutic use. The 8D4-8 antibody has been reported for use as a capture antibody for human IL-4 ELISA and for intracellular staining for flow cytometric analysis.
This tandem dye is sensitive to photo-induced oxidation. Protect this reagent
from light during storage and all handling steps. When staining cells with
PE-Cy7-conjugated antibodies, always protect samples from light.
Applications Tested
The fluorochrome-conjugated 8D4-8 antibody is offered in 2 formats:
- μg size: has been tested by intracellular staining for flow cytometric analysis. This can be used at less than or equal to 0.25 μg per million cells in a 100 μl total staining volume. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
- test size: has been pre-titrated and tested by intracellular staining for flow cytometric analysis. This can be used at 20 μl (0.125μg) per million cells in a 100 μl total staining volume.
