Staining of non-transfected (left) and mouse TLR3 transfected (right) 293 cells with purified rat IgG isotype control (open histogram) or 1:1000 diluted rat serum anti-mouse TLR3 (colored histogram) followed by biotin anti-rat IgG (13-4813) and Streptavidin-PE (12-4317). Total viable cells were used for analysis.

Description

The TLR3 rat polyclonal antibody reacts with mouse TLR3 and was generated using the sequence of extracellular domain of mouse TLR3. To date, at least twelve members of the Toll family have been identified. This family of type I transmembrane proteins is characterized by an extracellular domain with leucine-rich repeats and a cytoplasmic domain with homology to the type I IL-1 receptor. In the innate immunity system, TLRs recognize molecular patterns associated with microbial pathogens and induce antimicrobial immune responses. TLR3 recognizes double-stranded (ds)RNA and induces the activation of NF-κB, through MyD88-dependent and -independent pathways and the production of type I interferons (IFNs). Immunoblotting with this polyclonal gives a MW of ~120 kDa in mTLR3 transfected cells.

Applications Reported

For research use only, not for diagnostic or therapeutic use. This rat serum has been reported for use in flow cytometric analysis and immunoblotting (WB) of TLR3 on transfected cells.

Applications Tested

Rat serum anti-mouse TLR3 has been tested by WB of mTLR3 cell lysate. This polyclonal serum can be used at a dilution of 1:500 for immunoblotting. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
