DescriptionThe mouse TLR2 polyclonal antibody reacts with mouse TLR2 and was generated using a peptide sequence corresponding to residues 296-321 (fnpsesdvvselgkvetvtirrlhipq). To date, at least twelve members of the Toll family have been identified in human and mouse. This family of type I transmembrane proteins is characterized by an extracellular domain with leucine-rich repeats and a cytoplasmic domain with homology to the type I IL-1 receptor. Two of these receptors, TLR2 and TLR4, are pattern recognition receptors and signaling molecules in response to bacterial lipoproteins and have been implicated in innate immunity and inflammation. TLR2 mRNA is expressed in mouse spleen, lung, smooth muscle, adipose tissue and to a lesser degree in kidney and heart. At the protein level, TLR2 is best detected by flow cytometry on myeloid cells in spleen using monclonal antibodies including 6C2, mTLR2.4 and mTLR2.7. Immunoblotting with this polyclonal gives a MW of ~97 kDa in RAW 264.7 cells. In some cell lysates a non-specific lower MW at ~50 kDa is also observed. It is reported that TLR2 expressed on resident peritoneal cells can be downregulated by treatment of mice with Thioglycolate, however an overnight stimulation of thioglycolate-elicited PEC with LPS results in upregulation of TLR2 on these cells. Applications ReportedFor research use only, not for diagnostic or therapeutic use. Rabbit serum anti-mouse TLR2 has been reported for use in immunoblotting (WB). Applications TestedRabbit serum anti-mouse TLR2 has been tested by WB of RAW 264.7 cell lysate. This polyclonal serum can be used at a dilution of 1:1000 for immunoblotting. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. References
