Source: Insect cells infected with baculovirus: amino acids met 1-gly 214 corresponding to the extracellular domain TIM-1 (accession #AF399830) fused with Xa cleavage site -human Fc-His6.
Molecular Mass: The intact protein corresponding to the extracellular TIM-1 domain (tyr 22 – gly 214) –Xa cleavage site-human Fc-His6 has a predicted molecular mass of 51,446. The DTT reduced protein migrates as a 65 kDa polypeptide on SDS-PAGE due to glycosylation. Some 51 kDa polypeptide is also present. On non-reducing SDS-PAGE the homodimeric cystine-linked protein migrates as a doublet at 95-120 kDa.
Purity: Greater than 98% as determined by SDS-PAGE
Endotoxin Level: Less than 0.01 ng/ug cytokine as determined by the LAL assay.

Description
The Tim-1-Fc is a soluble fusion protein derived from expression of a fusion of cDNA encoding the entire extracellular domain of mouse Tim-1 to a cDNA encoding a human IgG1 Fc tail. TIM-1, a member of newly identified T cell immunoglobulin domain, mucin-like domain (TIM) gene family. TIM family of proteins are transmembrane proteins expressed by CD4 T cells. Recent studies demonstrate that the TIM family, particularly TIM-1, plays a critical role in immune responses that regulate the development of atopic diseases (e.g., asthma, allergic rhinitis, atopic dermatitis). Human TIM-1 is a functional receptor for hepatitis A virus (HAV). In humans, certain polymorphic variants of TIM-1 are strongly associated with protection against atopy, and this association occurs only in individuals who have had past infection with hepatitis A virus (HAV). Activation of T cells through TIM-1 by HAV or by its natural ligand may affect T cell differentiation and the development of Th2-driven allergic inflammatory responses.
Applications Reported
