Description


The 2D6 monoclonal antibody reacts with mouse CD22, which is a member of the Siglec (sialic acid-binding immunoglobulin-like lectin) family. CD22 is a 150 kDa type I membrane protein expressed by the B-cell lineage in a developmentally-regulated pattern. In mouse, CD22 is expressed at low levels on the surface of pre-B cells and immature (B220loIgMhi) B cells, but high levels on mature (B220hiIgMintIgD+) B cells in the bone marrow. In the periphery, mouse CD22 is highly expressed on all B-cell subsets. Furthermore, mouse CD22 expression is up-regulated slightly upon activation, however expression is lost upon differentiation to a plasma cell. CD22 plays a role in the negative regulation of B-cell Receptor (BCR) signaling, however it has also been demonstrated to function as an intercellular adhesion molecule through binding of sialic acid-bearing proteins expressed by lymphocytes, neutrophils, monocytes and non-hematopoietic cells.


Applications Reported


For research use only, not for diagnostic or therapeutic use. This 2D6 antibody has been reported for use in flow cytometric analysis.


Applications Tested


This 2D6 antibody has been tested by flow cytometric analysis of mouse splenocytes. This can be used at less than or equal to 0.5 μg per million cells in a 100 μl total staining volume. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
