Staining of human Fn14-transfected L5178Y cells with 0.06 μg of Purified Mouse IgA Iso Cntrl (cat. 14-4762) (open histogram) or 0.06 μg of Purified anti-hu/mo Fn14 (ITEM-2) (colored histogram) followed by FITC anti-mouse IgA (cat. 11-4204). Total viable cells were used for analysis.

Description


The ITEM-2 antibody reacts with human TWEAK Receptor/Fn14 (fibroblast growth factor-inducable 14 kDa protein) and cross-reacts with the mouse antigen. Fn14 is distantly related to the TNFR family, containing one cysteine-rich domain in the extracellular region and a TNFR-associated factor binding domain but does not contain a death domain (DD) cytoplasmic region. Fn14 plays a role in TWEAK-induced endothelial cell migration, proliferation, and angiogenesis. TWEAK-induced cell death via Fn14 includes both apoptosis and necrosis and can be blocked by an anti-TWEAK antibody, CARL-1 (Cat. No. 16-9915). Fn14 is expressed on HUVEC and in some cancer tissues but not on freshly isolated PBMCs. Fn14 mRNA expression has been identified during liver regeneration. In the mouse, Fn14 is reported to be constitutively expressed by Colon 26 cell line. ITEM-2 is reported to act as an antagonist in in vitro functional assays.


Applications Reported


For research use only, not for diagnostic or therapeutic use. The ITEM-2 antibody has been reported for use in flow cytometric analysis. It has also been reported in in vitro blocking of mouse/human Fn14. (Please use Functional Grade purified ITEM-2, cat. 16-9016, in functional assays.)


Applications Tested


The ITEM-2 antibody has been tested by flow cytometric analysis of human Fn14 transfected cells. This can be used at less than or equal to 0.125 μg per million cells in a 100 μl total staining volume. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
