Staining of human c-Met/GFP-transfected cells with 0.5 μg of Purified Rat IgG1 Iso Cntrl (cat. 14-4301) (left) or 0.5 μg of Purified anti-human c-Met (eBioclone 97) (right) followed by PE Anti-Rat IgG (cat. 12-4822). Total viable cells were used for analysis.

Description


The eBioclone 97 monoclonal antibody was generated against a human c-Met-Ig fusion protein, and reacts with human c-Met (HGFR)-transfected cells. Human c-Met is a 145 kDa receptor tyrosine kinase (RTK) expressed by epithelial cells of the brain, kidney, liver and other tissues. Binding of its ligand, Hepatocyte Growth Factor (HGF), triggers receptor autophosphorylation, and activation of several downstream effectors including the mitogen-activated protein kinases ERK-1 and ERK-2, and PLC-γ. Activation of the c-Met signal transduction pathway leads to mulitple cellular responses including cell motility, scattering, proliferation, survival and angiogenesis. Mutations in human c-Met have been implicated in the development of several malignancies.

The eBioclone 97 monoclonal antibody has been tested by flow cytometric analysis of human c-Met-transfected BAF/3 cells. The low level of expression of c-Met in primary tissue may make flow cytometric detection of the endogenous c-Met protein difficult. Experiments are underway to evaluate use eBioclone 97 for flow cytometric detection of c-Met from primary cells. Please contact eBioscience technical support for further information.

Applications Tested


This eBioclone 97 antibody has been tested by flow cytometric analysis of human c-Met-transfected cells. This can be used at less than or equal to 1 μg per million cells in a 100 μl total staining volume. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
