Staining of mouse c-Met-transfected 293T cells with 0.25 μg of Purified Rat IgG1 Iso Cntrl (cat. 14-4301) (open histogram) or 0.25 μg of Purified anti-mouse c-Met (eBioclone 4) (colored histogram) followed by Biotin Anti-Rat IgG (cat. 13-4813)and SAv-PE (cat. 12-4317). Total viable cells were used for analysis.

Description

The eBioclone 4 monoclonal antibody was generated against a mouse c-Met-Ig fusion protein, and reacts with mouse c-Met (HGFR)-transfected cells. Mouse c-Met is a 170 kDa receptor tyrosine kinase (RTK) expressed by epithelial cells of the brain, kidney, liver and other tissues. Binding of its ligand, Hepatocyte Growth Factor (HGF), triggers receptor autophosphorylation, and activation of several downstream effectors including the mitogen-activated protein kinases ERK-1 and ERK-2, and PLC-γ. Activation of the c-Met signal transduction pathway leads to mulitple cellular responses including cell motility, scattering, proliferation, survival and angiogenesis. Mutations in human c-Met have been implicated in the development of several malignancies.

The eBioclone 4 monoclonal antibody has been tested by flow cytometric analysis of mouse c-Met-transfected 293T cells. The low level of expression of mouse c-Met in primary tissue, including bone marrow cells from adult mice, may make flow cytometric detection of the endogenous c-Met protein difficult. Experiments are underway to evaluate use eBioclone 4 for flow cytometric detection of c-Met from primary cells. Please contact eBioscience technical support for further information.

For research use only, not for diagnostic or therapeutic use. This eBioclone 4 antibody has been reported for use in flow cytometric analysis.

Applications Tested

This eBioclone 4 antibody has been tested by flow cytometric analysis of mouse c-MET-transfected 293T cells. This can be used at less than or equal to 1 μg per 100 μl blood (or per 1 million cells in 100 μl total staining volume). It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
