Formulation: Sterile liquid; 10 mM sodium phosphate, pH 7.2, 150 mM NaCl, with 0.5% BSA. 0.22 μM filtered.
Storage Conditions: For greatest stability, keep concentration of primary stock at or above 10 µg/ml. For long term storage, aliquot into polypropylene vials (volumes of 20 µl or greater) and store at or below -80°C. Avoid repeated freeze/thaw cycles.
Handling Conditions: For best recovery, always quick-spin vial prior to opening. For dilution of current stock, always include carrier protein (1% BSA or 10% FBS) in the buffered saline diluent.
Source: E. coli expressed amino acids Cys 24-Asp 189 of mature human IFNα1 (accession # NM_024013).
Molecular Mass: The protein is not methionylated at the N-terminal and has a predicted molecular mass of 19,386. The DTT reduced protein migrates as a 21 kDa polypeptide on SDS-PAGE and the non-reduced protein migrates as a 16 kDa polypeptide due to intramolecular disulfides.
Purity: Greater than 98% as determined by SDS-PAGE
Endotoxin Level: Less than 0.01 ng/ug cytokine as determined by the LAL assay.
Bioactivity: The recombinant human IFN-α1 has been tested for inhibition of the cytopathic effect of EMC virus on A549 cells. The ED50 for this effect is typically below 0.02 ng/ml, corresponding to a specific activity of 5 x 107 U/mg.

Description


Human IFN-α1 is a type I interferon previously known as B-cell interferon, leukocyte interferon, lymphoblast interferon, and pH2-stable interferon. IFN-α1 is one of at least 23 different known variants of IFN-α. The individual proteins have molecular masses between 19-26 kDa and consist of proteins with lengths of 156-166 and 172 amino acids. The IFN-α forms are produced by monocytes/macrophages, lymphoblastoid cells, fibroblasts, and other cell types following induction by viruses, nucleic acids, glucocorticoid hormones, and low-molecular weight substances (n-butyrate, 5-bromodeoxyuridine). Human IFN-α1 promotes antiviral, antiparasitic, and antiproliferative activities.

Applications Tested
