Formulation: Sterile 10 mM NaH2PO4, 150 mM NaCl, 1 mM DTT, with 0.5% BSA. 0.22 µm filtered.
Storage Conditions: For greatest stability, keep concentration of primary stock at or above 10 µg/ml. For long term storage, aliquot into polypropylene vials (volumes of 20 µl or greater) and store at or below -80°C. Avoid repeated freeze/thaw cycles.
Handling Conditions: For best recovery, always quick-spin vial prior to opening. For dilution of current stock, always include carrier protein (1% BSA or 10% FBS) in the buffered saline diluent.
Source: E. coli expressed amino acids His 23-Cys 155 of mouse IFN-γ (accession # NM_008337).
Molecular Mass: The protein is methionylated at the N-terminal. The DTT reduced and protein migrates as a 14.5 kDa polypeptides on SDS-PAGE.
Purity: Greater than 98%, as determined by SDS-PAGE
Endotoxin Level: Less than 0.01 ng/ug cytokine as determined by the LAL assay.
Bioactivity: Measured by mouse L929/EMCV cell protection assay. The ED50 is 100 pg/ml, corresponding to a specific activity of 1.0 x 107 Units/mg.

Description


Mouse IFN-γ is a ~20 kDa factor produced by activated T, B and NK cells, and is an anti-viral and anti-parasitic cytokine. IFN-γ, in synergy with other cytokines such as TNF-α, inhibits proliferation of normal and transformed cells. Immunomodulatory effects of IFN-γ are exerted on a wide range of cell types expressing the high affinity receptors for IFN-γ. Glycosylation of IFN-γ does not affect its biological activity.


Applications Reported


For research use only, not for diagnostic or therapeutic use. Recombinant mouse IFN-γ is biologically active and can be used as a standard in mouse IFN-γ ELISA.


Applications Tested


This reagent has been tested by sandwich ELISA using the anti-mouse IFN-γ ELISA antibody pair (XMG1.2/R4-6A2 or AN-18/R4-6A2) and in bioassays using the mouse fibroblast cell line L929. The ED50 measured in a L929 protection is typically 100 pg/ml, corresponding to a specific activity of approximately 1x107 Units/mg.
