Sizes: 5 ug, 25 ug
Formulation: Sterile liquid; 0.1M Glycine, pH 3.0 with 1.0% BSA. 0.22 µm filtered.
Storage Conditions: For greatest stability, keep concentration of primary stock at or above 10 µg/ml. For long term storage, aliquot into polypropylene vials (volumes of 20 µl or greater) and store at or below -80°C. Avoid repeated freeze/thaw cycles.
Handling Conditions: For best recovery, always quick-spin vial prior to opening. For dilution of current stock, always include carrier protein (1% BSA or 10% FBS) in the buffered saline diluent.
Source: E. coli expressed protein consisting of amino acids Ser 24-Thr 164 of mature human IL-31 (accession # NM_001014336).
Molecular Mass: The DTT reduced protein migrates as a 15 kDa polypeptide on SDS-PAGE.
Purity: Greater than 97%, as determined by SDS-PAGE
Endotoxin Level: Less than 0.01 ng/ug cytokine as determined by the LAL assay.
Bioactivity: Measured by induction of STAT3 activation in U-87 cells. 5 ng/mL of rhIL-31 can effectively induce STAT3 activation.

Description
Human Interleukin-31 (IL-31) is a T-cell derived cytokine. The human IL-31 cDNA encodes a 164 amino acid precursor that contains a 23 amino acid signal peptide and a 141 amino acid mature protein. Human and mouse IL-31 share 24% amino acid sequence identity in the mature region. IL-31 is mainly associated with activated T cells and preferentially expressed by Th2 rather than Th1 cells. IL-31 signals via a heterodimeric receptor complex composed of a 120 kDa, gp130-like molecule termed IL-31RA (also GPL and GLM-R) and a 180 kDa oncostatin M receptor (OSM Rb). GPL/OSMR signaling is a strong activator of STAT3 and STAT5, and can also activate STAT1, Jak1, and Jak2 signaling pathways. IL-31 signaling has been shown to involve the Jak/STAT pathway, the PI3 kinase/AKT cascade, and MAP kinase pathway. The IL-31 receptor is constitutively expressed by keratinocytes and upregulated by IFN-g on monocytes.
Applications Reported
For research use only, not for diagnostic or therapeutic use. Purified Recombinant human IL-31 has been reported for use in cytokine bioassays.
Applications Tested
This recombinant human IL-31 has been tested by bioassays using the cell line U-87.
