Formulation: Sterile liquid; 10 mM sodium citrate, pH3.0, with 1.0% BSA.
Storage Conditions: For greatest stability, keep concentration of primary stock at or above 10 µg/ml. For long term storage, aliquot into polypropylene vials (volumes of 20 µl or greater) and store at or below -80°C. Avoid repeated freeze/thaw cycles.
Handling Conditions: For best recovery, always quick-spin vial prior to opening. For dilution of current stock, always include carrier protein (1% BSA or 10% FBS) in the buffered saline diluent.

Molecular Mass: The protein is methionylated at the N-terminal and has a predicted molecular mass of 15,135 daltons. The DTT reduced protein migrates as a 15 kDa polypeptide. The nonreduced cystine linked protein migrates as a 30 kDa protein on non-reducing SDS-PAGE.
Purity: Greater than 96% as determined by SDS-PAGE
Endotoxin Level: Less than 0.01 ng/ug cytokine as determined by the LAL assay.
Bioactivity: Measured by induction of IL-6 production by NIH/3T3 cells. The ED50 is 150 ng/ml, corresponding to a specific activity of 6.6 x 103 Units/mg.

Description
Rat IL-17A (IL-17), also known as CTLA-8, is a proinflammatory cytokine member of a six-species family of proteins (IL-17A-17F). Rat IL-17A protein is a homodimer consisting of two 134 amino acids peptides. IL-17A is secreted mainly by activated CD4+ and CD8+ T lymphocytes and acts through its receptor, IL-17R, to induce the expression of many mediators of inflammation, more importantly, those that are involved in the proliferation, maturation and chemotaxis of neutrophils. Elevated levels of IL-17A have been associated with several conditions, including rheumatoid arthritis, airway inflammation, allograft rejection, inflammatory bowel disease, psoriasis, cancer and multiple sclerosis. There is 58% identity between the amino acid sequence of human and rat IL-17A. Recombinant rat IL-17A produced in E.coli is a non-glycosylated disulfide-joined homodimer having a molecular mass of 30 kDa.
Applications Reported
For research use only, not for diagnostic or therapeutic use. Recombinant rat IL-17A is biologically active and can promote IL-6 production in vitro. Recombinant rat IL-17A is useful in ELISA assay as well.
Applications Tested
The recombinant rat IL-17A has been tested in a mouse IL-17 sandwich ELISA and in bioassay for induction of IL-6 production by NIH/3T3 cells. The ED50 is 150 ng/ml, corresponding to a specific activity of 6.6 x 10E3 Units/mg.
