Formulation: Sterile liquid; phosphate buffered saline, pH 7.2, 150 mM NaCl, with 1.0% BSA. 0.22 µm filtered.
Storage Conditions: For greatest stability, keep concentration of primary stock at or above 10 µg/ml. For long term storage, aliquot into polypropylene vials (volumes of 20 µl or greater) and store at or below -80°C. Avoid repeated freeze/thaw cycles.
Handling Conditions: For best recovery, always quick-spin vial prior to opening. For dilution of current stock, always include carrier protein (1% BSA or 10% FBS) in the buffered saline diluent.
Source: E. coli expressed amino acids ser 19-ser 178 of mature mouse IL-10 (accession # NM_010548) with a cys149tyr substitution (Moore, K. et al., Science 248:1230-1234).
Molecular Mass: The protein does not contain an N-terminal methionine. The polypeptide has a predicted molecular mass of approximately 18,950. The DTT reduced protein migrates as a 19 kDa polypeptide. The nonreduced protein migrates as an 18 kDa protein on non-reducing SDS-PAGE.
Purity: Greater than 98%, as determined by SDS-PAGE
Endotoxin Level: Less than 0.01 ng/ug cytokine as determined by the LAL assay.
Bioactivity: Measured by mouse MC/9 cell proliferation assay, costimulated with IL-4. The ED₅₀ is 0.3 ng/ml, corresponding to a specific activity of 3.0 x 10⁶ Units/mg.

Description

Mouse IL-10 is an ~18 kDa cytokine, also known as Cytokine Synthesis Inhibitory Factor (CSIF). In the mouse, Th2 cells, B1 cells and keratinocytes are the major cell subsets that produce IL-10. IL-10 inhibits synthesis of Th1 cytokines and proliferation of T cells, and acts as a costimulatory signal for mast cells, developing thymocytes and the Th2 response.

Applications Reported

For research use only, not for diagnostic or therapeutic use. Recombinant mouse IL-10 is biologically active and can promote proliferation of the MC/9 cell line in culture. It is also used as a cytokine standard in mouse IL-10 ELISA.

Applications Tested

This recombinant mouse IL-10 has been tested by sandwich ELISA using the mouse IL-10 ELISA antibody pair (JES5-16E3/JES5-2A5) and in bioassays using the mouse cell line MC/9. The ED₅₀ measured is typically 300 pg/ml, corresponding to a specific activity of 3.3x10⁶ Units/mg.
