C57BL/6 splenocytes were surface-stained with APC anti-mouse CD3ε, clone 145-2C11 (cat. 17-0031). Subsequently, cells were fixed and permeabilized, stained with 0.5 μg of FITC anti-mouse SLP-76 (clone MS76) (right) or mouse IgG2a isotype control (cat. 11-4724) (left) and analyzed by flow cytometry.

Description


The MS76 antibody reacts with mouse SLP-76. SLP-76 (SH2 domain-containing leukocyte protein of 76 kD) has also been described as lymphocyte cytosolic protein 2 (LCP2 ) and 76-kDa phosphoprotein (pp76 ). SLP-76 is a cytosolic adaptor protein expressed in several cell types of hematopoietic origin. SLP-76 associates with a number of additional molecules and is required for coupling receptor ligation with more distal signaling events in immature thymocytes, mature T cells, platelets, and mast cells. SLP-76 was originally identified as a substrate of the ZAP-70 protein tyrosine kinase following T cell receptor (TCR) ligation in the leukemic T cell line Jurkat. A number of additional proteins have been identified that associate with SLP-76 both constitutively and inducibly following receptor ligation, supporting the notion that SLP-76 functions as an adaptor or scaffold protein. Studies using SLP-76 deficient T cell lines or mice have provided strong evidence that SLP-76 plays a positive role in promoting T cell development and activation as well as mast cell and platelet function.

For research use only, not for diagnostic or therapeutic use. The MS76 antibody has been reported to be useful for IP, Western, and intracellular staining for flow cytometric analysis.


Applications Tested


The affinity purified format of this antibody has been tested by IP (1-5 ug) and Western blotting (1 ug/ml). The fluorochrome labelled MS76 antibody has been tested by intracellular staining with flow cytometric analysis of fixed and permeabilized mouse splenocytes.
