IL-1RA is produced by macrophages, monocytes, neutrophils, synovial and dermal fibroblasts. The 17 kDa soluble form of IL-1RA (sIL-1RA) is produced by hepatocytes. IL-1RA inhibits the release of both forms of IL-1, the secretion of IL-2, and the expression of IL-2 receptors on the cell surface. IL-1RA also blocks the stimulation of prostaglandin E2 synthesis in synovial cells and the proliferation of thymocytes. Applications ReportedFor research use only, not for diagnostic or therapeutic use. The CRM17 antibody has been reported for use in intracellular staining for flow cytometric analysis, and ELISA capture. Applications TestedThe CRM17 antibody has been tested as the capture antibody in a sandwich ELISA for analysis of human IL-1 receptor antagonist, in combination with the biotinylated rabbit anti-human IL-1RA antibody (13-7014) as the detection antibody and recombinant human IL-1RA as the standard. A suitable range of concentrations of this antibody for ELISA capture is 8-12 µg/ml. A standard curve consisting of doubling dilutions of the recombinant standard over the range of 10,000 pg/ml - 80 pg/ml should be included in each ELISA plate. The fluorochrome conjugated CRM17 antibody has been tested by intracellular staining and flow cytometric analysis.
