Staining of C57Bl/6 splenocytes with 0.5 μg of Purified Rat IgG2a Iso Cntrl (cat. 14-4321) (open histogram) or 0.5 μg of Purified II/41 (colored histogram) followed by Biotin Anti-Rat IgG (cat. 13-4813)and SAv-PE (cat. 12-4317). Total viable cells were used for analysis.

Description


The II/41 monoclonal antibody reacts with the μ heavy chain of mouse IgM. It does not react with other classes of mouse immunoglobulin including IgD, IgG or IgA. IgM is expressed intracellularly, during early stages of B lymphopoiesis, and then on the surface of more mature B cells in the bone marrow and peripheral B cells. Fluorochrome conjugated II/41 can be used as a detection secondary for mouse IgM.


Applications Reported


For research use only, not for diagnostic or therapeutic use. The II/41 antibody has been reported for use in flow cytometric analysis, immunohistochemical staining of frozen tissue sections, and ELISA.


Applications Tested


The II/41 antibody has been tested by flow cytometric analysis of mouse spleen cell suspension. This can be used at less than or equal to 1 μg per million cells in a 100 μl total staining volume. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
