Staining of normal human peripheral blood platelets with 0.06 μg of Purified Mouse IgG1, K Iso Cntrl (cat. 14-4714) (left) or 0.06 μg of Purified anti-human CD61 (VI-PL2) (right) followed by PE Donkey Anti-Mouse IgG (H+L) (cat. 12-4012). Samples were then extensively washed and co-stained with FITC anti-human CD42b (HIP1) (cat. 11-0429) to label platelets. CD42b(-) events represent cell debris.

Description
The VI-PL2 monoclonal antibody reacts with human CD61, also known as integrin β3 and GPIIIa. CD61 is a 90-110 kDa member of the /beta integrin family expressed by a wide variety of cells, including leukocytes, platelets, endothelial and smooth muscle cells. CD61 binds non-covalently with the α integrins CD41 and CD51, to form the αIIbβ3 (CD41/CD61) and αvβ3 (CD51/CD61) complexes. Theseαβ heterodimers are capable of mediating a variety of cellular responses including adhesion, trafficking, proliferaton and differentiation. The CD41/CD61 integrin has been shown to be involved in platelet aggregation, and binds to fibrinogen, von Willebrand Factor (vWF) and fibronectin. The CD51/CD61 integrin binds to matrix proteins including vitronectin, fibronectin, vWF and fibrinogen, and has been shown to have a strong role in modulating the migration and survival of angiogenic endothelial cells.
Applications Reported
For research use only, not for diagnostic or therapeutic use. This VI-PL2 antibody has been reported for use in flow cytometric analysis, immunoprecipitation, immunohistochemical staining, and functional assays.
Applications Tested
This VI-PL2 antibody has been tested by flow cytometric analysis of human peripheral blood platelets. This can be used at less than or equal to 0.125 μg per 100 μl blood (or per 1 million cells in 100 μl total staining volume). It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
