Staining of BALB/c splenocytes with 0.25 μg Purified Rat IgG2a Iso Cntrl (cat. 14-4321) (open histogram) or 0.25 μg Purified anti-mouse CD16/32 (93) (colored histogram) followed by FITC Anti-Rat IgG (cat. 11-4811). Total viable cells were used for analysis.

Description


The 93 monoclonal antibody reacts with an epitope shared by mouse CD16 and CD32. CD16 (Fcγ III Receptor) and CD32 (Fcγ II Receptor) are the low affinity receptors for the mouse IgG Fc portion and are expressed by B cells, monocyte/macrophages, NK cells, and neutrophils.


Applications Reported


For research use only, not for diagnostic or therapeutic use. The 93 antibody has been reported for use in flow cytometric analysis. 93 has also been reported in blocking of Fc-mediated reactions in functional studies. For blocking of Fc receptors in flow cytometric analysis, pre-incubate the cells with 0.5-1 µg of purified anti-CD16/CD32 per million cells for 5-10 minutes on ice prior to staining. (Please use Functional Grade purified 93, cat. 16-0161, in functional assays.)


Applications Tested


The 93 antibody has been tested by flow cytometric analysis of mouse splenocyte suspensions. This can be used at less than or equal to 0.5 μg per million cells in a 100 μl total staining volume. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
