Human peripheral blood mononuclear cells were stimulated with PMA and ionomycin in the presence of Brefeldin A for 5 hours. The cells were surface stained with FITC anti-human CD4 (RPA-T4) and intracellularly stained with PE anti-human TNF-a (Mab11).

Description


The MAb11 antibody reacts with human tumor necrosis factor-alpha (TNF-α ), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells and CD4+ T cells. TNF-α has cytolytic activity against a range of tumor cells and is important in immune regulation. TNF-α forms dimers and trimers and also exists as a 26 kDa membrane bound form. The MAb11 antibody is a neutralizing antibody.


Applications Reported


For research use only, not for diagnostic or therapeutic use. The MAb11 antibody has been reported for use in detection of human TNF-α by ELISA.


Applications Tested


The MAb11 antibody has been tested as the detection antibody in a sandwich ELISA for analysis of human TNF-alpha (TNF-a, TNFa) in combination with the purified MAb1 (14-7348) antibody for capture and recombinant human TNFa (14-8329) as the standard. A suitable range of concentrations of this antibody for ELISA detection is 0.5-2 µg/ml. A standard curve consisting of doubling dilutions of the recombinant standard over the range of 1000 pg/ml - 8 pg/ml should be included in each ELISA plate.
