Description

This biotinylated F(ab)'2 prepared from an antigen-affinity purified polyclonal rabbit antibody reacts with mouse IL-13. This antibody is useful as the detection antibody in a mouse IL-13 sandwich ELISA.

Mouse interleukin-13 (IL-13), originally designated p600, is a 11.6 kDa protein secreted by activated T helper cells, CD8+ T cells, and NK cells. IL-13 was originally identified in 1989 through differential hybridization of cDNA libraries prepared from T helper 1 and T helper 2 cells. IL-13 contains four cysteine residues, which form two intramolecular disulfide bonds. IL-13 shares many activities with IL-4; this relates to a requirement for IL-4Ralpha chain for signal transduction. The biological activities of IL-13 include suppression of macrophage cytotoxic activity, upregulation of IL-1RA expression, and suppression of inflammatory cytokine expression. IL-13 induces CD23 expression on B cells and enhances CD72, and expression of class II MHC gene products. Mouse IL-13 does not display the activities on T cells and B cell stimulatory effects which IL-4 exhibits. Mouse IL-13 is active on human and mouse cells.

Applications Reported

For research use only, not for diagnostic or therapeutic use. The biotinylated F(ab)'2 of an antigen-affinity purified fraction of a polyclonal rabbit antibody has been reported for use as the detection antibody in sandwich ELISA for analysis of mouse IL-13 protein levels, in combination with the purified eBioMTZ4513 antibody as the capture antibody.

Applications Tested

The biotinylated F(ab)'2 of an antigen-affinity purified fraction of a polyclonal rabbit antibody has been tested as the detection antibody in a sandwich ELISA for analysis of mouse interleukin-13 (IL-13) in combination with the purified eBioMTZ4513 (14-7131) antibody for capture and recombinant mouse IL-13 (14-8131) as the standard. A suitable range of concentrations of this antibody for ELISA detection is 0.25 - 1.0 µg/ml. A standard curve consisting of doubling dilutions of the recombinant standard over the range of 4000 pg/ml - 30 pg/ml should be included in each ELISA plate.
