Staining of FC-blocked (anti-CD16/32, cat. 14-0161) BALB/c peritoneal-exudate cells with FITC anti-mouse/human CD11b (M1/70) (cat. 11-0112) and 0.25 μg of Biotin Armenian Hamster IgG Iso Cntrl (cat. 13-4888) (left) or 0.25 μg of Biotin anti-mouse CCR5 (HM-CCR5) (right) followed by SAv-PE (cat. 12-4317). Total viable cells were used for analysis.

Description
CCR5 is the major co-receptor for macrophage-tropic isolates of HIV-1. CCR5, also known as CD195, is a seven transmembrane chemokine receptor capable of binding RANTES, MIP-1α and MIP-1β. Knockout mice show a decreased recruitment of macrophages and increased susceptibility to infection. Protein and mRNA has been found in macrophages from thioglycolate-treated and activated T cells.
The HM-CCR5(7A4) monoclonal antibody binds the N terminal extracellular domain of mouse CCR5.
Applications Reported
For research use only, not for diagnostic or therapeutic use. This HM-CCR5 (7A4) antibody has been reported for use in flow cytometric analysis.
Applications Tested
This HM-CCR5 (7A4) antibody has been tested by flow cytometric analysis of mouse thioglycolate elicited peritoneal cells (PECS). This can be used at less than or equal to 0.5 μg per million cells in a 100 μl total staining volume. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
