Human PBMCs were surface-stained with APC anti-human CD8 (RPA-T8) (cat. 17-0088) and FITC anti-human CD56 (MEM188) (cat. 11-0569) then fixed and permeabilized and subsequently with PE anti-human Perforin (delta G9) or PE mouse IgG2b Iso Cntrl (cat. 12-4732). The density plot on the left demonstrates co-staining of CD56 and delta G9 while the right plot demonstrates co-staining of CD8 and delta G9. Quadrant lines demarcate the isotype control. Cells in the lymphocyte gate were used for analysis.

Description

The dG9 antibody clone reacts with human perforin (pore-forming protein, pfp). Perforin is one of the cytolytic mediators present in the cytoplasmic granules of cytotoxic T lymphocytes (CTL) and natural killer cells (NK). Perforin is involved in the killing function by CTLs and NKs and has an important role in the immune response against tumors and virus infections.

Applications Reported

For research use only, not for diagnostic or therapeutic use. This dG9 (delta G9) antibody has been reported for use in intracellular staining followed by flow cytometric analysis.

Applications Tested

This dG9 (delta G9) antibody has been pre-titrated and tested by intracellular staining and flow cytometric analysis of human PBMCs. This can be used at 20 μl (0.125 μg) per 100 μl blood (or per 1 million cells in 100 μl total staining volume).
