Staining of 24-hour LPS activated normal human peripheral blood mononuclear cells with 0.25 μg of PE Armenian Hamster IgG Iso Cntrl (cat. 12-4888) (open histogram) or 0.25 μg of PE anti-mo/hu/rat CCL2/MCP-1 (2H5) (colored histogram). Cells in the monocyte gate were used for analysis.

Description

The 2H5 monoclonal antibody reacts with mouse, rat, and human monocyte chemoattractant protein-1 (MCP-1), also known as CCL2 and MCAF.

Applications Reported

For research use only, not for diagnostic or therapeutic use. The 2H5 antibody has been reported for use in ELISA, intracellular staining for flow cytometric analysis, and neutralization. (Please use Functional Grade purified 2H5 antibody, cat. 16-7096, in functional assays.)

Applications Tested

This 2H5 antibody is offered in 2 formats:

- μg size: has been tested by flow cytometric analysis of fixed permabilized LPS-stimulated thioglycolated mouse PEC cells. This can be used at less than or equal to 0.25 μg per 100 μl blood (or per 1 million cells in 100 μl total staining volume). It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

- test size: has been pre-titrated and tested by intracellular staining and flow cytometric analysis of LPS-stimulated normal human peripheral blood mononuclear cells. This can be used at 20 μl (0.25 μg) per 100 μl blood (or per 1 million cells in 100 μl total staining volume). The pre-titrated test size contains 0.2% BSA.
