Detection of Human IL-4 Producing Cells
(L): Human Peripheral blood mononuclear cells were stimulated with PMA and ionomycin in the presence of 2 μM monensin for 5 hours. The cells were then surface-stained with PE anti-human CD3 (UCHT1) and intracellular-stained with FITC anti-human IL-4 (MP4-25D2).
(R): The specific staining was confirmed by blocking assay through pre-incubation of the permeabilized cells with non-labeled purified anti-human IL-4 (MP4-25D2) before the intracellular staining.

Description
The MP4-25D2 antibody reacts with human interleukin-4 (IL-4), a 15-19 kDa cytokine secreted by Th2 cells.
Applications Reported
For research use only, not for diagnostic or therapeutic use. MP4-25D2 has been reported for use in intracellular flow cytometric analysis.
Applications Tested
The MP4-25D2 antibody is offered in 2 formats:
- µg size: has been tested by intracellular flow cytometric analysis and can be used at less than or equal to 0.5 μg/million cells (100 μl blood). It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
- test size: has been pre-titrated and tested by intracellular flow cytometric analysis and can be used at 20 μl (0.25μg) per million cells (100 μl blood). The pre-titrated test size contains 0.2% BSA.
