Jurkat cells treated for 4 hours (left) and 24 hours (right) with 10µM camptothecin were stained with the Annexin V-FITC Apoptosis Detection Kit (88-8005) and 5µl of Propidium Iodide (PI) Staining Solution in 100µl total staining volume. The Annexin V/ PI double-positive cells are late stage apoptotic and necrotic cells present after the 24 hour camptothecin (topoisomerase I inhibitor) treatment, but mostly absent from the 4 hour treatment.

Description
Propidium Iodide (PI) is a ready-to-use solution for the exclusion of nonviable cells in flow cytometric analysis. PI binds to double stranded DNA by intercalating between basepairs, but is excluded from cells with intact plasma membranes. PI can be used in FL3 for inviability exclusion, but should be analyzed in FL2 when used as a counterstain for FITC Annexin V.
Applications Reported
For research use only, not for diagnostic or therapeutic use. Propidium Iodide can be used as a viability probe for methods of nonviable cell exclusion, based on light scatter and uptake of the reagent as detected in FL3 or FL2.
CAUTION: PI is a potential carcinogen. It is recommended that the user wear protective clothing, gloves, and eye/face protection in order to avoid contact with skin and eyes.
Applications Tested
Propidium Iodide Staining Solution has been pre-titrated and tested on Jurkat cells treated with 10uM camptothecin for 24 hours. This can be used at 5 μl per million cells in a 100 μl total staining volume.
