beta-Catenin is a component of the WNT/Frizzled pathway. Beta-Catenin associates with the cytoplasmic domain of E-Cadherin, a protein that helps govern cell-cell adhesion, as well as the tumor suppressor adenomatous polypopsis coli (APC). GSK3 phosphorylates both beta-Catenin and APC, thereby marking them for degradation. Wnt binding to the Frizzled receptor induces GSK3 phosphorylation, which leads to inhibition of its kinase activity and subsequent reduction in ß-Catenin phosphorylation and degradation. As such, beta-Catenin translocates into the nucleus and functions as a co-activator of the TCF/LEF transcription factor family, which plays key roles in regulating genes involved in development and cancer progression.
